Tm value is the temperature at which 50% of the base pairs of a DNA duplex are separated. This Tm value depends on the sequence length, GC-binding rate, and concentration.
There are several methods to calculate Tm values. As an example, we could supply the “Nearest Neighbor method” on an Excel sheet available for calculation.
